乳胶微球在免疫诊断学方面有着非常广泛的应用。采用乳胶微球可以从血清、尿和脑脊液中检测出微量的抗原或者抗体。微量的抗体可以使抗原包被的微球产生明显的聚集;同样微量的抗原也可以使抗体包被的微球产生一样明显的聚集。

Latex microspheres have a very wide range of applications in immunodiagnosis.Latex microspheres can detect trace amounts of antigens or antibodies in serum,urine, and cerebrospinal fluid. Micro amounts of antibodies can causesignificant aggregation of antigen-coated microspheres; The same trace amountof antigen can also cause similar aggregation of antibody coated microspheres.

另外一种较常见的用途是将其应用在乳胶凝集抑制试验中。将待测样品中的抗原与已知抗体作用后,再与相应抗原包被的乳胶微球混合。因没有游离抗体的存在,乳胶颗粒表面的抗原不能与抗体结合出现凝集现象,即凝集被抑制。

现有羧基/氨基聚苯乙烯微球、羧基/氨基聚甲基丙烯酸甲酯微球和表面空白的聚苯乙烯微球三种微球。其中羧基/氨基聚苯乙烯微球和表面空白的聚苯乙烯微球的材质为聚苯乙烯高聚物,并用二乙烯基苯交联,形成网状聚合物结构;以及羧基/氨基聚甲基丙烯酸甲酯微球的材质为聚甲基丙烯酸甲酯。产品为水分散体系,有很好的力学性能、耐溶剂、耐各种pH环境,以及表面富含羧基/氨基和硫酸根,在溶液中可以保持很好的稳定性,可以适用于各种缓冲溶液。微球表面光滑、平整,比表面积大,并具有很好的均一性、单分散性和稳定性。此类羧基/氨基微球可以较容易地将羧基/氨基与蛋白进行共价偶联,而表面空白的聚苯乙烯微球可以与蛋白通过疏水缔合作用进行连接。在免疫比浊检查、免疫荧光检测、ELISA增强、乳胶凝集试验等领域有着广泛的应用。

There arethree kinds of microsphere: carboxyl/amino polystyrene microsphere,carboxyl/amino polymethyl methacrylate microsphere and polystyrene microspherewith blank surface. The carboxyl/amino polystyrene microspheres and the surfaceblank polystyrene microspheres are made of polystyrene high polymer andcrosslinked with divinylbenzene to form a network polymer structure; And thecarboxyl/amino polymethyl methacrylate microspheres are made of polymethylmethacrylate. The product is a water dispersible system with excellentmechanical properties, solvent resistance, and resistance to various pHenvironments. The surface is rich in carboxyl/amino groups and sulfateradicals, which can maintain good stability in the solution and can be appliedto various buffer solutions. The surface of the microspheres is smooth andflat, with a large specific surface area and good uniformity, monodispersity,and stability. These carboxyl/amino microspheres can easily covalently couplecarboxyl/amino groups with proteins, while surface blank polystyrenemicrospheres can be connected to proteins through hydrophobic association. 1免疫荧光检测

免疫荧光检测的原理是先将抗体固载于微球表面,负载于微球表面的抗体与抗原特异性结合后,再加入荧光标记的第二抗体与抗原结合,使微球-抗体-抗原-第二抗体-荧光标记复合物具有稳定的荧光信号,经过洗涤后,用仪器检测荧光信号。荧光信号的强度与披测抗原的浓度有较强的相关性。

1. Immunofluorescence detection

The principle of immunofluorescence detection is to firstimmobilize antibodies on the surface of microspheres, and then add a secondantibody labeled with fluorescence to bind to the antigen. 2胶乳颗粒增强免疫比浊法

胶孔颗粒增强免疫比浊法 (PETIA) 分为两种。一种是散射比浊检测法:另一种是透射比浊检测法。这两种方法的基本原理都是在高分子胶孔微球的表面交联抗体或抗原,当交联有抗体或抗原的微球与抗原或抗体结合后,在短时间内会迅速聚集在一起,改变了反应液的散光性能或透光性能。而且,反应液散光性能或透光性能(即吸光度)的改变与被测抗原或抗体的浓度有较强的相关性,在一定范围内可以反映被测物的浓度

2 latex particles enhanced immunoturbidimetry

3 ELISA反应的增强

此类应用是基千ELISA反应的原理,使用粒径范围在 60-200nm的乳胶微球,用酶标仪进行检测。区别在于传统的ELISA反应采用了酶标抗体,而此应用是将酶和抗体一起偶联到微球的表面,使一个微球表面连接有多个酶分子和抗体分子,将偶联有大昼酶分子和抗体分子的微球代替传统的一个酶分子标记一个抗体的酶标抗体对ELISA反应起到一个增强的作用。

Enhanced ELISA response

This type of application is based on the principle of ELISAreaction, using latex microspheres with a particle size range of 60-200nm fordetection using an enzyme-linked immunosorbent assay (ELISA). The difference isthat traditional ELISA reactions use enzyme-linked antibodies, while thisapplication couples enzymes and antibodies to the surface of microspheres,connecting multiple enzyme molecules and antibody molecules on the surface of amicrosphere. 4乳胶凝集法

乳胶凝集法是以乳胶颗粒作为载体的一种间接;肚集试验。即吸附或偶联可溶性抗原或抗体千其表面,特异性抗体或抗原与之结合后,使该乳胶颗粒发生清晰凝集。凝集反应在玻瑶板上进行,结果在良好的光照下用肉眼观察,常用千类风湿因子检查等方面。

4 latex agglutination method
